Resumen

The objective was to evaluate the effect of caffeine (CAFN) and Rhus trilobata (RT) on oocyte maturation. Cumulus oocytes complexes (COC’s) were aspired from cows ovaries provided from a slaughterhouse, cultivated for 23 h, and distributed in four groups: CONTROL, CAFN 10 mM, RT 3 µg / mL, and CAFN+RT. Compaction of COC’s, polar body (CP) extrusion, ROS – GSH levels, development of oocytes after parthenogenesis, and In vitro fertilization rate were evaluated. The COC's were maintained compacted (64.30 %) and meiotic resumption was kept inhibited by action of CAFN; only just a low percentage extruded CP at the end of the maturation (5.77 %). The RT improved nuclear maturation (CP; 42.81 %) in comparison with the rest of the groups and kept the COC’s compacted. The CAFN increased (P < 0.05) ROS contents (1.6539 UAF) in comparison with the RT (1.1805 UAF) and CAFN+RT (1.0815 UAF). GSH concentration was similar (P > 0.05) among treatments. The celular division of the oocytes artificially activated or fertilized was similar (P > 0.05) among groups. These results suggest that the RT promotes the ability of develop competition of the oocyte whereas the CAFN acted as a potent meiosis inhibitor, besides it decreases oocyte viability and ability to develop competition of the oocyte.
