Resumen

In vitro embryo production of white-tailed deer (VCB) is not well documented. The objective of this study was to conduct maturation (IVM) and in vitro fertilization (IVF) using VCB oocytes vitrified with Trehalose (TH) or Sucrose (SC). The oocytes were vitrified in two groups: 1) TH (n = 60) and 2) SC (n = 61). The oocyte warming (OW) was done, then the viability evaluation in TH (n = 5) and SC (n = 5) and the nuclear status (NS) in TH (n = 4) and SC (n = 5) were evaluated by classifying them into Germinal Vesicle (GV), Metaphase I and Not evaluable (NE). IVM was performed for 36 h in supplemented TCM-199 and mature oocytes were evaluated (n = 88) for NS [GV, MI = immature, MII = mature]. For IVF, frozen semen with a concentration of 3 x 106 sperm / ml was used, and the incubation was for 24 h in supplemented TALP medium. At this stage, the NS was seen as fertilized (F), unfertilized (NF) or NE. For the statistical analysis an exact Fisher test was used, α = 0.05. For the TH group (n = 5), after warming, a viability of 60% was obtained, while for the SC (n = 5) the viability was 40%. All the oocytes were immature. In the IVM stage, maturation is not shown for the TH group (n = 38) and for the SC group (n = 50) only 2% of mature oocytes. In the IVF stage, with TH (n = 10) no fertilization occurred while with SC (n = 5) 20% fertilization was obtained. However, no difference (P> 0.05) was found between the TH and SC groups. Due to the reduced sample, future research with a bigger sample of immature or mature oocytes is suggested for further evaluation of both TH and SC for WTD oocyte vitrification for use with IVP techniques as a model for other endangered cervid species.
