Resumen

Intermedin/Adrenomedulin2 (IMD/ADM21-47) is an oocyte secretion peptide that inhibits apoptosis and promotes interactions in the cumulus-oocyte complex (COCs) in rats. The objective was to evaluate the impact of adding the IMD/ADM21-47 to the medium culture for bovine oocyte maturation, with the perspective to be considered as a promoter of competition in bovine oocytes. To evaluate the effect on cell compaction of COCs, nuclear maturation and apoptosis into the oocyte, groups of 50 COCs were matured with 15, 153 and 510 µg/ml of IMD/ADM21-47 for 24 h. COCs cultivated at 510 µg/ml showed a more compact structure (88% ± 16.32 P <0.05) and those cultivated at 153 µg/ml had a higher percentage of mature oocytes (61% ± 3.8 P <0.05) and less apoptosis (2% ± 2.3, P <0.05) than the rest of the treatments. There was no effect on the expression of the bax and blc2 genes, so the anti-apoptotic effect of IMD/ADM21-47 probably involves the extrinsic pathway of apoptosis. To measure the effect on cAMP production, groups of 50 COCs were matured for 0, 3, 6 and 24 h in 153 µg/ml IMD/ADM21-47, 3 μM Forskolin (FSK) as a positive control, and IMD/ADM21-47+FSK. At 0 h, the cAMP was higher in IMD/ADM21-47+FSK (38.37, P <0.05) than in the rest of the treatments. At 6 h, cAMP concentrations were higher in the treatment of IMD/ADM21-47 (70.2 fmol/mL ± 2.3, P <0.05); and at 24 h in FSK treatment (48.9 fmol/mL ± 1.05, P <0.05). In conclusion, supplementation with the peptide IMD/ADM21-47 to the culture medium for in vitro maturation has a beneficial impact, as it promotes cell-cell interaction by maintaining the compact structure in the COCs, which favors the synthesis of cAMP to improve at MII stage and inhibition of apoptosis in the oocytes.
