Resumen

The aim of this study was to determine the effects of adding Rhus trilobata plant extract (RT, 16.6 mg/mL) to the bovine semen extender compared to Cysteine (CIS, 2 mM/mL), Caffeine (CAF, 5 mM/mL) and Ascorbic Acid (AA, 4.5 mg/mL) on seminal post-thawing traits. Two experiments were conducted. In experiment 1, semen of six bulls was processed adding to the extender: No additive (Control, CT), CIS, CAF or RT. After thawing, viability, plasma membrane functionality (IFMC), motility and capacitation status were assessed. Experiment 2, was performed to evaluate the intracellular concentration of glutathione (GSH) in sperm cells in response to the addition of CIS or RT to semen extender in cryopreservation of bulls younger than 2 years old and older. The data for both experiments were analyzed under a mixed model with SAS using PROC MIXED. In experiment 1, the addition of the RT improved (P < 0.05) viability and IFMC (4 and 8%, respectively) compared to CT. Total motility was increased (P < 0.05) by RT, CIS and AA. Higher (P < 0.05) progressive motility was observed with RT, AA and CT. The spermatic kinematics was lower (P < 0.05) in velocity characteristics in the AA group. However, RT treated cells tend to show higher values. Greater percentage (P < 0.05) of sperm no capacitated with intact acrosome was obtained with CT, CIS and RT. In the second experiment results show increased (P < 0.05) levels of glutathione in bulls older than 2 years in response to RT extract. In conclusion, addition of RT extract can improve post-thawing quality of bovine cryopreserved semen
